Calibration, standards and matrix effects
Quantitative analysis needs a validated relation between response and analyte amount.
Key knowledge
Analytical calibration establishes an instrument or method response using standards. An external calibration assumes the sample and standards have a compatible response relation. Matrix matching or validated standard additions can address some matrix effects; calibration alone does not establish selectivity.
Conditions and scope
- Use the validated response range, blanks and uncertainty assessment.
- Standard additions require an appropriate response model and comparable effects on native and added analyte; they do not correct every interference.
