Molecular barcodes and missing observations

UMIs help distinguish amplified reads from captured molecules, but barcode errors and finite capture affect estimated counts.
Key knowledge
UMIs help distinguish amplified reads from captured molecules, but barcode errors and finite capture affect estimated counts. A zero observed gene count is not automatically biological absence, and normalization cannot recover every uncaptured molecule.
Conditions and scope
- Barcode collision/error treatment, library depth and cell-size/composition differences matter; do not equate read count, UMI count and true molecular abundance.
