Perturbation, rescue and gene-function evidence
A genetic perturbation tests a causal prediction when the intended change and relevant phenotype are measured against appropriate controls.
Key knowledge
A genetic perturbation tests a causal prediction when the intended change and relevant phenotype are measured against appropriate controls. Agreement across independent perturbations or clones and restoration by a suitable resistant allele can strengthen gene-function attribution. DNA alteration, transcript level, protein detection and phenotype are different readouts; none automatically certifies all the others.
Conditions and scope
- Interpret results within the assayed cell type, state and environment. Rescue expression must be appropriate: overexpression or altered regulation can change the interpretation.
- Off-target and unintended on-target changes, clone variation, compensatory function and reagent specificity can confound conclusions. A missing antibody band can reflect loss of an epitope or an undetected functional fragment rather than complete protein loss.

Evidence and review
Read the primary method's critical-parameters/rescue and Understanding Results passages for clone, antibody and protein-fragment confounding; read the original second protocol abstract for unintended target-locus alterations escaping standard checks. This is evidence interpretation, not a laboratory procedure, clinical recommendation or assertion that a single rescue proves every mechanism.
